Background
The defined antibody specificity is to the hepatitis associated antigen as tested in counterelectrophoresis and double radial immunodiffusion against HAA positive human serum. The product does not react with normal human serum or plasma. In precipitating techniques as counteroelectrophoresis and double radial immunodiffusion (Ouchterlony) to identify the presence of hepatitis associated antigen.

The immunogen has been prepared from sera of patients with hepatitis using gel filtration Freund’s complete adjuvant is used in the first step of the immunization procedure.
Product
Delipidated, heat inactivated, lyophilized, stable whole serum. No preservative added. Total protein and IgG concentrations in the antiserum are comparable to those of normal pooled swine serum. No foreign proteins added. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water.

Specificity
Precipitating polyclonal Swine antiSerum to Human hepatitis associated antigen.
Species Reactivity: Inter-species cross-reactivity of this antiSerum has not been tested in detail.
Applications
Precipitation assays. In counter electrophoresis use opposite holes at a distance of 4mm with 6 μl antiserum and serum samples. In double radial immunodiffusion use a rosette arrangement with 10 μl antiserum in 3 mm diameter center well and 2 μl plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells. The distance between the wells should be 2-3 mm. In case the concentration of the protein component is significantly increased or reduced, the required equivalence between the reactants can be establish by increasing the amount of antiserum or antigen respectively.
Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
