Background
The reactivity of the antiserum is directed to the Fc portion of the subclass IgG1. It does not react with other subclasses of IgG, IgG/Fab fragments, IgM and IgA or any non-Ig protein in rat serum, as tested by immunoelectrophoresis and double radial immunodiffusion. In precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgG1 in rat serum and other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of rat IgG1 from serum or plasma.

Pools of purified homogenous IgG1 isolated from pooled rat serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Product
Delipidated, heat inactivated, lyophilized, stable whole antiserum. No preservative added as it may interfere with he antibody activity. Total protein and IgG concentration in the antiserum are comparable to those of pooled normal sheep serum. No foreign proteins added. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water.

Specificity
Precipitating polyclonal Sheep antiSerum to Rat IgG1, subclass specific.
Species Reactivity: This IgG fraction is not species-specific since inter-species cross-reactivity is a normal feature of antisera to immunoglobulins. Cross-reactivity of this product has not been tested in detail, however in double radial immunodiffusion a reaction with Mouse has been obtained.
Applications
Precipitation assays. In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl antiserum in a 3 mm diameter centre well and 2 μl serum samples (neat and diluted) in 2 mm diameter peripheral wells.
Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
