Sheep anti Human salivary amylase

Background
The reactivity of the antiserum is restricted to salivary amylase. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against human saliva a single precipitin line is obtained and shows a reaction of full identity with the purified immunogen. No reaction is obtained with human plasma. In precipitating techniques as immunoelectrophoresis and single and double radial immunodiffusion (Mancini, Ouchterlony) to identify the presence of salivary amylase or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibodybinding techniques if proper controls are included.
Source
Human salivary amylase is an enzyme hydrolyzing the alpha-1,4-glucan links in polysaccharides. Its molecular weight is 55,000. The immunogen has been isolated from pooled saliva and purified to homogeneity for use in the antiserum production. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Product
Delipidated, heat inactivated, lyophilized, stable antiserum. No preservative added Total protein and IgG concentration are comparable to those of pooled normal sheep serum. No foreign proteins added Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water.
Formulation: Delipidated, heat inactivated, lyophilized, stable antiserum. No preservative added Total protein and IgG concentration are comparable to those of pooled normal sheep serum. No foreign proteins added
Specificity
Precipitating polyclonal Sheep antiSerum to Human salivary amylase.
Species Reactivity: The antiSerum does not cross-react with any other Human plasma proteins as tested in gel-diffusion techniques. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins, since homologous proteins of different species frequently share antigenic determinants. Cross-reactivity of this antiSerum has not been tested in detail.
Applications
Precipitation assays. In immunoelectrophoresis in agarose-plates use 2 μl human saliva or equivalent against 120 μl antiserum. In double radial immunodiffusion use a rosette arrangement with 10 μl antiserum in 3 mm diameter center well and 2 μl plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells.
Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
