Background
The reactivity of the antiserum is directed to the Fc subunits of IgE as tested against rat sera and a panel of purified homogenous immunoglobulins. It does not react with IgG, IgG/Fab fragments, IgM and IgA or any non-Ig protein in rat serum, as tested by immunoelectrophoresis and double radial immunodiffusion. In precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of homogenous IgE in rat serum and other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of rat IgE from serum or plasma.

Pools of purified homogenous IgE isolated from pooled rat serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Product
Delipidated, heat inactivated, lyophilized whole antiserum. No preservative added, as it may interfere with the antibody activity. Total protein and IgG concentration in the antiserum are comparable to those of pooled normal rabbit serum. No foreign proteins added. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water.

Specificity
Precipitating polyclonal Rabbit antiSerum to Rat IgE, Fc specific.
Species Reactivity: This IgG fraction is not species-specific since inter-species cross-reactivity is a normal feature of antisera to immunoglobulins. Cross-reactivity of this product has not been tested in detail because of the low level of IgE in most species.
Applications
Precipitation assays. In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl antiserum in a 3 mm diameter centre well and 2 μl serum samples (neat and diluted) in 2 mm diameter peripheral wells.
Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
