Background
The reactivity of the antiserum is restricted to subclass specific determinants on IgG3. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against normal human serum a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified IgG3. No precipitation reaction is obtained with purified other subclasses of IgG, IgA, IgM, and IgG/Fab fragments.

Highly purified homogenous IgG3 isolated from human serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Product
Delipidated, heat inactivated, lyophilized, stable whole antiserum. No preservative added. Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal rabbit serum. No foreign proteins added. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water.

Specificity
Precipitating polyclonal Rabbit antiSerum to Human IgG3, subclass specific.
Species Reactivity: The antiSerum does not cross-react with any other component of the Human immunoglobulin system. Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Cross-reactivity of this antiSerum has not been tested in detail.
Applications
Precipitation assays. In double radial immunodiffusion (Ouchterlony) serum or plasma samples are prepared in serial two-fold dilutions from 1:8 to 1: 128 and placed in the peripheral wells of the rosette arrangement. One well is filled with pooled human serum or another control. The center wells are filled with the subclass specific antiserum and the sera are allowed to diffuse at ambient temperature for 16-24 hours. Frequently the precipitin lines become visible only after staining.
Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
