Background
The major histocompatibility complex (MHC) is a cluster of genes that play an important role in the immune response. In humans, this complex is referred to as the Human Leucocyte Antigen (HLA) system and is located on chromosome 6. In humans the MHC II locus includes genes for alpha and beta subunits of the three class II MHC proteins HLA-DR, HLA-DQ and HLA-DP. The HLA class II alpha and beta genes are highly polymorphic with the exception of HLA-DR alpha gene, which is reported to be monomorphorphic. HLA class II glycoproteins are expressed as alpha/beta heterodimers on the surface of antigen presenting cells such as dendritic cells, B-cells and monocytes. Their primary function is to present foreign antigen derived peptides to CD4 positive T-cells. HLA-class II molecules also play an important role in developing and maintaining tolerance to self-antigens and in transplant rejection. HLA-DR molecules have been associated with a range of autoimmune diseases including rheumatoid arthritis, Grave’s disease and multiple sclerosis.

Bra30 is a mouse monoclonal IgG2a antibody derived by fusion of mouse myeloma cells with cells from a mouse immunized with REH-6 cells, a human pre-B cell leukaemia cell line.
Product
Each vial contains 500 µl 0.2 mg/ml purified monoclonal antibody in phosphated buffered saline (PBS) containing 50% glycerol + 0.2% bovine serum albumin (BSA) + 0.02% sodium azide.

Specificity
The antibody Bra30 is directed against a monomorphic determinant of human HLA Class II. The antibody recognises an epitope on the HLA-DR molecule that is dependent on the association of both heavy and lights chains of the HLA class II molecule. The antibody does not cross react with either HLA-DP or HLA-DQ molecules. The antibody has not been tested for cross reactivity with other species.
Applications
The Bra30 antibody is suitable for the detection of HLA Class II antigen by flow cytometry, immunocytochemistry and immunohistochemistry on frozen tissues. The antibody is also suitable for the detection of HLA class II by immunoprecipitation and western blotting.
Optimal antibody dilutions for the different applications should be determined by titration.
Storage
The antibody is shipped at ambient temperature and may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
