Background
Confirmed by immunoelectrophoresis and double radial immunodiffusion (Ouchterlony). Not less than 98% as determined by agar-gel electrophoresis, quantitative densitometry immunoelectrophoresis and double radial immunodiffusion (Ouchterlony). The ratio of IgA subclasses IgA1 and IgA2 has been determined and found to be 60/40. This secretory IgA contains J chain and secretory component. The product contains a trace of non-Ig protein.
Source
Human colostrum.
The protein is isolated according to a method described in J. Mestecky and M. Kilian, Immunoglobulin A (IgA), in Methods in Enzymology, Vol 116, pp 37-75, 1985
Product
Chromatographically purified Human secretory IgA.
Lyophilized, stable white powder. No preservative added The lyophilized content is reconstituted by adding 0.5 ml of sterile distilled water, mixed gently by inversion until complete dissolution is obtained. When applied as an sIgA standard the exact concentration of the dissolved product must be estimated before use. An extinction coefficient of 1.26 should be used for a 1 mg/ml solution of sIgA.
Applications
Purified human secretory IgA may be used as a reference antigen, calibrator, coating protein and blocking agent in a variety of immunoassays including immunodiffusion, immunoelectrophoresis, ELISA, Western blotting, dot-immuno-binding assays (DIBA), haemagglutination, and cell-binding assays.
Storage
This product is shipped lyophilized at ambient temperature. After arrival store at +4°C. Prolonged storage may be at -20°C. Allow to stand at ambient temperature for 5-10 minutes to reach equilibrium. Reconstituted solution may be stored frozen. Working dilutions may be prepared by adding the required amount of phosphate buffered saline (PBS), should not be refrozen and preferably used within 24 hours. Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation and will not affect the performance of the product.
Shipping Conditions: Ship at ambient temperature
