Background
The reactivity of the antiserum is restricted to the Fc part of the IgA molecule. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against normal rabbit serum a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified IgA. No precipitation reaction is obtained with purified IgG, IgM, and IgG/Fab fragments. In precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgA in rabbit serum or other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of rabbit IgA from serum or plasma. This antiserum is not intended for use in non-precipitating antibody-binding or other highly sensitive assays. For such use labelled and unlabelled cytochemical and immunoassay grade reagents meeting the necessary additional specificity and performance requirements are available.

Highly purified normal IgA isolated from rabbit serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Product
Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added. It is reconstituted by adding 1 ml sterile distilled water, spun down to remove insoluble particles, divided into small aliquots, frozen and stored at or below -20°C.

Specificity
Precipitating polyclonal Goat antiSerum to Rabbit IgA, Fc specific.
Species Reactivity: The antiSerum does not cross-react with any other component of the Rabbit immunoglobulin system. Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Cross-reactivity of this antiSerum has not been tested in detail.
Applications
Precipitation assays. In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum. In double radial immunodiffusion use a rosette arrangement with 10 μl antiserum in a 3 mm diameter centre well and 2 μl serum samples (neat and diluted) in 2 mm diameter peripheral wells.
Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Prior to use, an aliquot is thawed slowly at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the product.
